Proline is a non-essential amino acid the body makes on its own and also obtains from dietary protein. It is a major building block of collagen.

Why it’s used

Taken by some as a standalone supplement to support skin, joint, or connective tissue health, often alongside other collagen-related amino acids like glycine and hydroxyproline.

What the evidence says

Proline's structural role in collagen is well established biochemically, but evidence that supplementing with free proline (as opposed to consuming dietary protein or collagen peptides) changes skin or joint outcomes is limited. There is no NIH Office of Dietary Supplements fact sheet specific to proline supplementation.
